Overview of Services

 

The Analytical Chemistry/Small Molecule Corefacility provides analytical chemistry services in support of clinical and basic science research within and outside of the Research Institute at Nationwide Children’s Hospital. Liquid and gas chromatography with mass spectroscopy capabilities are available for monitoring and quantifying compounds of interest, with a particular focus on small molecules. Services will also be provided for the method development and validation of new assays.

 

Available Instrumentation


  • Liquid Chromatography- Mass Spec/Mass Spec (LC-MS/MS) currently equipped for small molecule analyses such as drugs, drug metabolites, and lipids.
  • Gas Chromatography-Mass Spec (GC-MS) equipped to analyze small molecules not amenable to liquid chromatography separation.
  • Gas Chromatography (GC) equipped with a flame ionization detector, currently used for fatty acid analysis.
  • High Pressure Liquid Chromatography two independent units available with UV/VIS, diode array, and fluorescence detection.
  • Multiplex analysis for protein molecules (ELISA based format) using an MSD platform, samples can be analyzed for chemokines, cytokines, and other signaling molecules.

Acetaminophen 

HPLC analysis with UV detection of Acetaminophen. Select metabolites can be included on a case-to-case basis.

 
Aspirin 

LC/MS/MS analysis of Aspirin and its active metabolite, Salicylic Acid.

 
Birth Control Hormones 

LC/MS/MS analysis of Etonogestral, 17-Medroxyprogesterone acetate, Norgestrel, and Progesterone.

 
Bosentan 

LC/MS/MS and LC/UV tandem analysis of the drug Bosentan and specific acid hydrolysis metabolites.

 
Cortisol 

Analysis for Cortisol in human hair or saliva using a competitive ELISA kit from Alpco.

 
Eicosanoids: plasma, breast milk, cells 

LC/MS/MS analyisis of a panel of oxygenated derivatives of arachidonic acid including select HETEs, DHETs, Eets, HODEs, Leukotrienes, Prostaglandins, Lipoxins, Resolvins, and Thromboxanes. Double charge for batch sets under 12 samples.

 
Eicosanoids: tissues 

LC/MS/MS analyisis of a panel of oxygenated derivatives of arachidonic acid including select HETEs, DHETs, Eets, HODEs, Leukotrienes, Prostaglandins, Lipoxins, Resolvins, and Thromboxanes. Double charge for batch sets under 12 samples. 

 
Ethylene Glycols 

LC/MS/MS analysis of ethylene glycol and its analogs, diethylene glycol and triethylene glycol, in plasma samples through dibenzoyl derivitization.

 
Fatty Acids by GC/FID 

GC/FID analysis of fatty acids ranging from C8:0 to C22:6 in a variety of biological and cell matrices including plasma, breast milk, amniotic fluid, etc.

 
Fatty Acids by GC/MS 

GC/MS analysis of select fatty acids found in low abundance, such as lignoceric acid (C24:0) and nervonic acid (C24:1), in biological and cell matrices.

 
Homocysteine 

HPLC analysis by fluorescence detection of Cysteine, Homocysteine, N-acetyl-cysteine, and Glutathione in plasma. Other sulfur containing analytes and other matrices will be considered.

 
Losartan 

LC/MS/MS analysis of the drug Losartan in plasma samples.

 
Nicotine, Cotinine, Caffeine 

LC/MS analysis for Nicotine, Cotinine, and Caffeine. Currently only validated in breast milk samples.

 
Nitric Oxide Pathway/Amino Acids 

HPLC with fluorescence detection for the amino acids included in the nitric oxide pathway: Arginine and metabolic by-products ADMA, SDMA, NMMA, Citrulline, and Ornathine. Select polyamines will be considered on a case-by-case basis. Double charge for batch sets under 12 samples.

 
Oxidation Markers - 8-OH dG plasma 

LC/MS/MS analysis of 8-hydroxy-2'-deoxyguanosine (8-OH-dG) in plasma. Double charge for batch sets under 12 samples. 

 
Oxidation Markers - 8-OH dG tissue 

LC/MS/MS analysis of 8-hydroxy-2'-deoxyguanosine (8-OH-dG) in tissues. Double charge for batch sets under 12 samples. 

 
Oxidation Markers - Nitrotyrosine plasma 

LC/MS/MS analysis of tyrosines including: 3-Cl-Tyrosine, 3-NO-Tyrosine (nitrotyrosine), o-Tyrosine, m-Tyrosine, and phenylalanine in plasma. Double charge for batch sets under 12 samples. 

 
Oxidation Markers - Nitrotyrosine tissues 

LC/MS/MS analysis of tyrosines including: 3-Cl-Tyrosine, 3-NO-Tyrosine (nitrotyrosine), o-Tyrosine, m-Tyrosine, and phenylalanine in tissues. Double charge for batch sets under 12 samples. 

 
Oxymetazoline 

LC/MS/MS analysis of the drug oxymetazoline.

 
Vitamin D 

LC/MS/MS analysis for Vitamin D metabolites, Vitamin D2 and D3. Double charge for batch sets under 12 samples. 

 
Vitamins A and E 

HPLC analysis by UV absorbtion of a-Tocopherol, g-Tocopherol, Retinol, Retinoic Acid, and 13-cis-Retinoic Acid in plasma samples. Double charge for batch sets under 12 samples.
