CRISPR (clustered regularly interspaced short palindromic repeats) is a gene editing technology. The ease and speed of using CRISPR have made the technology a toolbox for everything from basic discovery to clinical therapy, with applications in oncology, immunology, infectious diseases, and genetic diseases etc. Its precision and safety have convinced the FDA to approve several clinical trials using CRISPR in the US. CRISPR services out of campus are expensive, takes a long time to perform, and are less available for consultation. Thus, CRISPR Core at NCH in collaboration with OSU CRISPR shared source will provide investigators at NCH and OSU with affordable prices, expert personnel, and reasonable turnaround time.
we will provide services in CRISPR Core including:
- Consultation on the most suitable approach for CRISPR gene modification.
- Providing CRISPR recombinant vectors encoding CRISPR gene targeting components.
- Helping researchers to design highly efficient guide RNAs.
- DNA-free CRISPR gene editing (Cas9/RNP).
- Generating highly efficient knock-out cell lines and hard to manipulate primary cells including primary NK and T-cells.
- Site directed gene insertion such as tagged proteins and CARs in several cells through CRISPR mediated gene insertion.
- Transient gene silencing or transcriptional repression (CRISPRi)
- Transient activation of endogenous genes (CRISPRa)
- Base editing.
- Gene editing in iPSC and HSC.
- Generation of gene edited grafts for transplantation
- We also can contribute and provide expertise and services which are needed for investigators grant applications.
